癌症一向是困扰人类的一个难题，随着科技的进步，癌症治疗领域日新月异，出现了很多新型疗法，精准医疗更是开启了癌症治疗的新时代。癌细胞是由正常细胞经过系类变异后所产生的，具有无限分裂、难以控制、易于转移等特点。而病毒一般由DNA或RNA与蛋白质构成。病毒个体微小、结构简单，通过吸附与进入、基因表达、转录、翻译、核酸复制、装配与释放等复制行为，在宿主细胞内进行自我复制，并最终导致宿主细胞出现死亡、增生、转化等。<img data-s="300,640" data-type="png" src="http://mmbiz.qpic.cn/mmbiz_png/dx3KibwtDhvdGCmkOmQ9piaGFodicgTjhc0icdEZFsCQrVlDzUKrWvFibk6HrCnrKd9a0bHOqWZth8G5CKa9C7eXBicg/0?wx_fmt=png" data-ratio="0.568075117370892" data-w="639" /> 基于癌细胞与病毒的一些特征，结合基因工程手段，采用病毒来治疗癌症的相关研究如火如荼。其中的主角溶瘤病毒便是一类倾向于感染肿瘤细胞，同时在癌细胞里面能够大量繁殖，最终让肿瘤细胞裂解、破碎、死亡的一类病毒。研究用于溶瘤媒介的主要病毒有腺病毒、痘病毒、疱疹病毒、呼肠孤病毒等，其中一些疗法已得到批准上市，一些也已进入临床研究阶段。2017年，让我们来重点关注对抗癌症的4种病毒。疱疹病毒（Herpes:）生物技术巨头安进（Amgen）的溶瘤病毒药物Imlygic （talimogene laherparepvec）于2015年成为美国FDA首次批准的一种病毒治疗药物。Imlygic 是一种基因改良的活体溶瘤细胞疱疹病毒，主要用于治疗不能通过手术完全切除的黑色素瘤病灶。作为一种单一疗法Imlygic的表现并非非常之出色，安进已经把Imlygic与默沙东（Merck）的Keytruda联合使用并开展了临床试验。早期试验结果令人颇为满意，也促使推动安进启动了3期临床试验，目前该试验尚在进行之中。今年7月份，新基生物制药（celgene）重金押注于另一源于疱疹病毒的疗法，该疗法由新兴公司Oncorus开发，尚处于试验阶段。致力于开发下一代溶瘤病毒疗法的Oncorus公司完成了5700万美元的A轮融资。新基是此轮的主要投资者。柯萨奇病毒（Coxsackievirus）澳大利亚制药公司Viralytics正致力于开发一种源于柯萨奇病毒（Coxsackievirus）的疗法，其中柯萨奇病毒是引起普通感冒的一种病毒。疗法Cavatak已与几种其它的不同疗法联合使用，包括与抗癌药物Keytruda及另一种来自于百时美施贵宝（BMS）的检查点抑制剂Yervoy （ipilimumab）联合使用，此2种疗法皆已批准用于治疗黑色素瘤。Viralytics公司10月份所宣布的早期试验结果是令人满意的：与Keytruda联合治疗的10位患者中，7位患者的肿瘤缩小；与Yervoy联合治疗的18位患者中，14位患者的病情得以控制。腺病毒（Adenovirus）百时美施贵宝（BMS）于6月份与英国制药公司PsiOxus Therapeutics就PsiOxus 的溶瘤腺病毒疗法enadenotucirev的临床开发签署了一份排他性合作协议。这项交易向业界证明了百时美施贵宝对检查点抑制剂与抗癌病毒疗法联合使用所抱有的希望。尽管医学领域已采用免疫检查点抑制剂来治疗多种肿瘤，但是仍存在着远未满足的医疗需求。Enadenotucirev作为一种病毒能够选择性地在肿瘤细胞中进行复制，而不会在正常细胞中复制，在某些实体瘤的治疗中具有潜力。就2家公司达成的合作协议，PsiOxus正在开发的工程型腺病毒enadenotucirev与百时美施贵宝的肿瘤免疫疗法 Opdivo联合起来用于治疗晚期癌症患者，研究溶瘤病毒治疗技术与免疫检查点抑制剂在各类型肿瘤中的协同疗效，以期对癌症患者提供一个更加有效的治疗手段。根据双方协议，百时美施贵宝将一次性支付PsiOxus 1000万美元前期款，I期临床试验由PsiOxus负责开展。双方将展开独家合作来开发抗PD-1/PD-L1拮抗抗体与enadenotucirev的组合疗法。呼肠孤病毒（Reovirus ）位于加拿大的公司Oncolytics Biotech的病毒性疗法Reolysin正处于2期临床试验阶段，Reolysin以呼肠孤病毒（Reovirus ）为基础，用于治疗多种类型实体瘤，包括乳腺癌、前列腺癌、胰腺癌等。为了检验Reolysin与其它几种药物联合使用的疗效，公司Oncolytics Biotech已开展过几个相关试验。Reolysin是一种被修饰过的病毒，自然形式的病毒可以引起呼吸道疾病。虽然Oncolytics Biotech公司在Reolysin的研发道路上一直被所失望困扰，也曾导致公司股价暴跌，并最终导致首席执行官Brad Thompson的离去。但是公司就Reolysin所开展的试验却在压力中继续推进着，最近公司公布了一项新的组合试验：把与Reolysin与Keytruda联合起来用于治疗胰腺癌。其他一些研究小组也对咳嗽病毒及其在抗癌上的潜力兴趣十足。11月份伦敦一组科学家宣布在小鼠试验中，呼肠孤病毒似乎可以刺激机体自身免疫系统来杀死肝癌细胞、并可以清除丙型肝炎病毒（HCV），其中HCV是一种常见的导致肝癌的病毒。现在他们正在计划在人体中展开相关试验。 来源：生物探索<p style="font-size: 16px;
